Abstract:
The screening of diabetes complications may greatly benefit from the adoption of point-of-care devices for detecting diabetes biomarkers in human blood plasma and urine samples. Preeminent Methylglyoxal (MG) and Carboxymethyl-lysine (CML) levels lead to various diseases and diabetes complications. Therefore, MG and CML are considered to be the potential biomarkers of diabetes mellitus, which are produced in the Maillard reaction and involved in the formation of advanced glycation end-products. In this background, we have developed electrochemical biosensors for the detection of MG and CML in human blood plasma and urine samples. An enzyme-based electrochemical MG biosensor was developed with a linear range of 1.0 - 7.5 μM concentration, sensitivity of 1.02 mA μM-1, limit of detection of 0.21 μM, limit of quantification of 0.70 μM and response time less than 10 s. The developed sensor was validated with 350 clinical samples and it exhibited a 90% of correlation with ELISA data. In addition, an enzyme-free antibody-based electrochemical sensor for the detection of CML was developed and it exhibited a linear range of 10 ng mL-1 – 500 μg mL-1. To obtain more accuracy in detecting the complications of diabetes, one can adopt a simultaneous detection of both MG and CML analytes with the multi-probe channel system.
